人胸膜间皮瘤UGTA17基因序列分析

Analysis of UGTA17 gene sequence of Human pleural mesothelioma

摘要: 以胸膜间皮瘤和非胸膜间皮瘤病理组织为研究对象,分析UGT1A7 基因突变与胸膜间皮瘤发生的关系。对楚雄彝族自治州人民医院50例胸膜间皮瘤患者病理组织及7例非胸膜间皮瘤患者病理组织进行UGT1A7 基因片段分离、测序,并将测序结果与正常人UGT1A7 基因参考系列进行比对。结果显示,4例胸膜间皮瘤患者的UGT1A7 基因片段发生了突变,病例XL17、XL20、XL41都在UGT1A7基因234676872位点(扩增片段401位点)发生碱基突变,碱基C突变成碱基T;病例XL21在UGT1A7基因23467979位点(扩增片段病例508位点)发生碱基突变,碱基A突变成碱基C,总突变率为8.00%。非胸膜间皮瘤与正常参考序列比对,Z2在401位点上的的碱基C突变为T,突变率为14.29%。以上研究说明,胸膜间皮瘤的发生与UGTA1A7基因突变有关。

Abstract: The relationship between the mutation of UGT1A7 gene and pleural mesothelioma was analyzed in the pathological tissues of pleural mesothelioma and non-pleural mesothelioma.UGT1A7 gene fragments were separated and sequenced in the pathological tissues of 50 pleural mesothelioma patients and 7 non-pleural mesothelioma patients in the people's hospital of Chuxiong Yi autonomous prefecture,and the sequencing results were compared with the reference series of UGT1A7 gene in normal people.The results showed that four patients with pleural mesothelioma had mutations in the UGT1A7 gene segment,Case XL17,XL20 and XL41 all had base mutation at 234676872 place(amplification fragment 401) in UGT1A7 gene,and base C mutated into base T,with a total mutation rate of 8.00%.Case XL21 had A base mutation at 23467979 (amplified fragment at 508) in UGT1A7 gene,and base A mutated into base C,with a total mutation rate of 14.29%.The above studies indicate that the occurrence of pleural mesothelioma is related to UGTA1A7 gene mutation,but whether it is the direct cause of the occurrence of pleural mesothelioma remains to be further expanded sample study.
