摘要: 以NCBI数据库中牛等9种哺乳动物的IGF-1R的蛋白序列和基因序列为研究对象,利用生物信息学的相关软件和网站,综合分析它们的IGF-1R基因序列和蛋白序列的组成结构及功能的相关性。结果表明:9种动物IGF-1R的基因序列中除了山羊和印度水牛外,其他动物均存在启动子和加尾位点;9种动物均含有开放阅读框,可编码完整的多肽链。9种动物IGF-1R的蛋白序列中氨基酸均属于酸性氨基酸,并非全部都有信号肽;除了印度水牛,牛IGF-1R的蛋白序列与其他几种动物的一致性和相似性都是在25~35之间;二级结构均以无规则卷曲为主要结构;牛的IGF-1R蛋白序列三级结构与其他几种动物的差别比较大,但结构比较稳定,9种动物的IGF-1R蛋白序列三级结构预测的模型构象符合立体化学规则。本研究为进一步探讨IGF-1R的结构和功能及动物生长发育繁殖提供一定的参考依据。

Abstract: In order to understand the gene and protein structure of Insulin-like growth factor-1 receptor in Bos taurus and other animals, to know the differences between the various structures of IGF-1R and the corresponding functions of these animals, in this paper, the protein and gene sequences of 9 species of IGF-1R mammals in the NCBI database including B. taurus, Maylandia zebra, Capra hircus, Bubalus bubalis, Ovis aries, Sus scrofa, Equus caballus, Camelus dromedaries and Homo sapiens was used as research objects. Using the relevant software and website of bioinformatics, the correlation between the composition structure and function of the gene sequence and protein sequence of IGF-1R of these 9 animals is comprehensively analyzed. The results of the study indicate the IGF-1R gene sequences of that some of the 9 animal have promoters and tailing sites, and some do not exist; all contain open reading frames and can encode complete polypeptide chains. The amino acids in the protein sequences of 9 animal IGF-1Rs are acidic amino acids, and not all of them have signal peptides; except for the B. bubalis, the identity and similarity of the protein sequence of bovine IGF-1R with other animals is between 25~35; the evolutionary trees constructed by multiple sequences have differences, one is the difference in evolutionary years, and the other is that the IGF-1R protein sequences of these animals have homology; The secondary structure is mainly composed of random coils; the tertiary structure of bovine IGF-1R protein sequence is quite different from other animals, but the structure is relatively stable. The predicted model conformation of the tertiary structure of IGF-1R protein sequence of 9 animals is in accordance with the rules of stereochemistry. The research results have a better understanding of the structure and function of bovine IGF-1R, and provide a certain reference for further discussion on the structure and function of IGF-1R, animal growth and reproduction.
